Tagged FlyFos TransgeneOme Library - fosmid DNA constructs (Nov 1, 2022)

The Drosophila genome contains >13,000 protein-coding genes, the majority of which remain poorly investigated, for reasons such as the lack of antibodies or reporter constructs to visualise these proteins. To help address this, the Drosophila TransgeneOme project generated a genome-wide collection of engineered fosmid constructs (fTRG fosmid constructs).
The fTRG fosmid construct library consists of 10,000 GFP-tagged clones, comprising tagged genes and most of their regulatory information (Sarov et al., 2016). Each genomic fosmid construct contains a multi-epitope tag for generating a specific protein with a tag at its C-terminus, the majority with the 2XTY1-SGFP-V5-preTEV-BLRP-3XFLAG tag. These constructs can be used to generate transgenic Drosophila stocks for a variety of downstream applications.
The tagged fosmid constructs are created, maintained and distributed via the Max Planck Institute of Molecular Cell Biology and Genetics (MPI-CBG).

The VDRC does not store or distribute the fTRG fosmid constructs but enables you to search here for constructs of interest (e.g. via gene name, CG number etc) and find the link for requesting specific constructs via https://transgeneome.mpi-cbg.de/transgeneomics/public/projects.html.
A subset of these fTRG fosmid constructs were used to generate transgenic Drosophila stocks and are available to order from the VDRC: https://stockcenter.vdrc.at/control/library_ftrg.

General price increase (Mar 30, 2022)
Dear VDRC users,
VDRC would like to inform you about our general price increase effective April 1, 2022. This is the first increase since 2019 and reflects increased costs due to inflation.
VDRC is a non-profit research organization and our running costs and thus your fees are subsidized. Currently ~25% of the total running costs are funded by a grant from the Austrian Federal Ministry for Science and Research and the city of Vienna. The remaining costs must be recovered from user fees. We hope you understand that the price increase is necessary to ensure that we continue to reach this target, thus securing the longer term availability of our collection for the community.

For United States customers: New regulations for importing Drosophila (May 3, 2021)
New documents and practices for importing Drosophila into the United States were implemented by the USDA as of April 5th 2021.
To receive your imported packages from the VDRC, you will need a "Letter of No Permit Required” (LONPR).
For information on how to obtain this document see VDRC shipping information.
The LONPR document greatly simplifies the import procedure since it:
- may be used for an unlimited number of shipments,
- may be used to import strains from any country,
- has no expiration date.
Shipping via the old documents (Courtesy Permit and a "Letter of No Jurisdiction") will still be valid until the permit expiry date, but we recommend that you obtain a new “Letter of No Permit Required” as soon as possible.
We thank the BDSC and USDA for greatly simplifying the import procedures for the Drosophila research community!

New resource: Fly Extract is available to order (Jul 15, 2020)
Dear VDRC users,
Fly Extract, from OregonR adults, is an essential component for culturing of many insect cell lines derived from ovaries or imaginal discs.
The VDRC has been providing Fly Extract for a number of Vienna Biocenter research groups and now extends this service to the Drosophila community. The VDRC Fly Extract is produced using a protocol developed by the lab of Julius Brennecke (IMBA, Vienna), which has been successfully growing OSC cells using this extract since 2010 (Sienski et al., Cell 2012).

Update regarding VDRC shipping operations (May 10, 2020)
Dear VDRC users,
Whilst abiding by the COVID-19 lockdown and social distancing rules to ensure the safety of our staff, the VDRC team is continuing to work in shifts to ensure all our fly stocks are healthy. Stock maintenance is still our highest priority, but we are now able to take a further step towards normality by ramping up our shipping operations.
What changed?
From now on we are resuming our pre-COVID system of processing orders immediately they are received and shipping them as soon as possible. We still cannot guarantee shipment within 2 weeks but will do everything possible to minimize the turnaround time.By default, we will ship your order as soon as we have made it ready, however, we appreciate that not everyone is in the fortunate situation of being able to receive their order. Shipping operations resumed (Apr 07, 2020)

Due to COVID-19 lockdown measures, the VDRC team has had to make some operational changes, but the good news is that we have continued maintaining our full stock collection, so all our stock IDs are available for ordering.
Stock maintenance will continue to be our highest priority, but we are pleased to inform you that we are now able to start shipping again. This means that you can continue placing orders via our website as normal and they will be processed as soon as possible. We’ll do our best to minimize the turnaround time, but it is currently not possible to guarantee shipment within 2 weeks.

COVID-19 emergency update (Mar 13, 2020)
Dear VDRC users,
Due to reasons beyond our control relating to COVID-19 containment measures (customs closing, cancelled flights etc.) we regret to inform you that we will be unable to continue with our full service at the current time. This means that we will be unable to uphold our guarantee of shipping orders within 2 weeks and will not be able to refund you if your package is delayed. Shipping update regarding coronavirus (Feb 28, 2020)
Dear VDRC users,
Due to the spread of COVID-19, some destinations may not be able to receive deliveries by Fedex or DHL. To avoid that we send your package and it doesn’t get through, we ask that you please keep an eye on the situation in your area inform us straight away if you will be unable to receive your package. Sub-library lists of RNAi lines (GD, KK, shRNA) (Dec 02, 2020)
Dear VDRC users,
For ease of ordering of defined, focused sub-libraries, we have created many sub-library lists of RNAi lines (GD, KK, shRNA).
The 'RNAi Sub-library' section includes the following lists:
- human related genes (e.g.human gene orthologs sub-library, human disease-related orthologs sub-library)
- genes associated with specific signaling pathways (e.g. JAK-STAT, Notch and Toll signaling pathways)
- genes associated by biochemical and/or biological function (e.g. transcription factor genes, RNA binding genes, ubiquitin-related genes).
By clicking on each sub-library, the list of available lines for the associated genes will be displayed, enabling you to “add all to cart” or to select a subset.

New lines added - Heidelberg CFD CRISPR library (Sep 04, 2019)
We are very pleased to announce that The Heidelberg CFD CRISPR Library, generated by the laboratory of Michael Boutros at the German Cancer Research Center (DKFZ) and Heidelberg University, has been donated to the VDRC for distribution.
These lines enable loss-of-function mutations in essential or non-essential genes in a germline restricted, ubiquitous or tissue specific fashion:
- ~1600 sgRNA lines primarily targeting:
- kinases, phosphatases and transcription factors
- some fly orthologs of genes implicated in human pathologies
- 20 toolkit stocks
- UAS-Cas9 lines with a range of expression levels
- UAS-Cas9 combined with common Gal4 driver lines
- Lines for induction of Cas9 expression by FLP-out

New price scheme - as of 4th September 2019 (Sep 04, 2019)
The Vienna Drosophila Resource Center (VDRC) is part of the Vienna Biocenter Core Facilities (VBCF), a non-profit research company, publicly funded by the Austrian Federal Ministry of Education, Science and Research, and the City of Vienna via the Vienna Business Agency.
Up until now, fees for use of the resources have provided approximately 50% of the cost of VDRC services with the remaining 50% coming from VBCF funding. However, since Austria currently represents less than 2% of the total VDRC usage, (with US ~40%, Europe ~40% and Asia ~15%) we are required to increase the proportion of our running costs coming from user fees.
To achieve this, we will be implementing a new price scheme as of 4th September 2019 [see here for details].
The good news is that the fees for orders of 6 stocks or less have actually been reduced. When ordering 7 lines (the median number of lines ordered by VDRC customers last year), we have increased the price by 2%.
To offset the slightly increased price for larger orders, we have improved our service to guarantee that all orders up to 300 lines (previously 100) will qualify for fast shipment and will be dispatched within 2 weeks. This means that the total cost of your order may still be cheaper than before.

Stock Pruning List, RNAi lines (GD and KK libraries) - order by 15th Sep 2019 (Aug 15, 2019)
Dear VDRC users,
In order to streamline the VDRC stock collection we will be discarding nearly 3500 RNAi lines (3339 GDs and 122 KKs) [list].
We aim to keep 2 independent lines per target gene, each line with a different construct and different target sequence. We have prioritised keeping lines with least off targets. After pruning there will still be at least 2 remaining RNAi lines (GD, KK or shRNA) targeting the same CG as the discarded line.
Please check the list carefully and ensure that you order the lines you need for current or future experiments. These stocks will no longer be available for ordering after Sep 15th 2019, so be sure to order in good time - they will be gone forever after this date!

New stocks added from Ameres lab (Mar 06, 2018)
Dear VDRC users,
The group of Stefan Ameres (IMBA, Vienna) has donated several new stocks to the VDRC. Included are stocks which can be used as a tool in a groundbreaking technique called "mime-seq". Mime-seq enables Gal4-driven access to the miRNA repertoire of specific tissues and cell-types in flies. The method therefore enables researchers to profile microRNAs in their endogenous context at cellular resolution - a level of specificity and sensitivity which was previously not possible.

Import regulation for US and Canada (Dec 22, 2017)
Dear VDRC users,
As of March 21, 2017, import regulations for the US and Canada have been amended regarding the "TSCA certification". From now on the TSCA must be signed by the Importer (VDRC user). Next time you place an order you will receive an automatic email notification with an attached TSCA which you have to fill out, sign and send back to us.
We will include this document with each shipment to the US and Canada.

"NEW" and "SIMILAR" tags added in search results (Jan 25, 2017)
We have made some changes to the way search results are displayed:
We've added the tag "NEW" to resources which have been recently added to the VDRC collection, to help you clearly distinguish them in the search results table.
The tag "SIMILAR" will appear when you search for a specific term and we wish to show you related resources. As an example, with the search term "33277" the search results will display the primary hit first (VDRC_ID 33277) and below it all related lines with the tag "SIMILAR", in this case 2 further resources matching the same CG number. If the search is more general e.g. with the search term "*10125", there can be more than one primary result from the search. In such a case, one primary result and its SIMILAR lines will be listed first, followed by the next primary result and its SIMILAR lines, with each related group separated by a solid line. Currently it is not possible to sort the SIMILAR lines, nor to filter them by library type, but we're working on it.
If the results are too confusing, we would suggest trying again with a more specific search term!

We have modernised our homepage (www.vdrc.at), not only to give it a fresh look, but also to make it even more user-friendly and to incorporate some new features.These include:
- Highlighting the most recently added resources
- A regularly updated list of the most popular lines
- Addition of links to FlyBase Reference Reports showing when the use of a specific line has been cited in publications
- Improved search features
- Ability to store lines in the shopping cart when you log out

New stocks added from Harvey lab (May 11, 2016)
Dear VDRC users,
Control lines for the KK library, donated by Kieran Harvey (Peter MacCallum Cancer Centre, Melbourne, Australia) are now available. (For details see Vissers et al, Nat. Comm. (2016) : A Drosophila RNAi library modulates Hippo pathway-dependent tissue growth).
The collection consists of 5 lines which can be displayed by using the search term "harvey":
An RNAi-toolkit stock (VDRC_ID 60101) where the landing sites at 40D and 30B in the KK library host strain have been separated by meiotic recombination. This line contains Gal4-responsive UAS repeats but no functional RNAi coding sequence at 40D and no transgene insertion at 30B and enables researchers to easily test whether their genetic screen of interest will be affected by ectopic tiptop expression.
4 further KK lines were created by meiotic recombination to separate the insertions at 30B and 40D. These lines contain an insertion of the UAS-RNAi construct in the landing site at 30B only and no insertion at the 40D landing site.
VDRC_ID 111000 is the "30D only" version of VDRC_ID 107151 (CG8208, MBD-like)
VDRC_ID 111001 is the "30D only" version of VDRC_ID 104523 (CG4005, Yki)
VDRC_ID 111002 is the "30D only" version of VDRC_ID 106174 (CG12072, Wts)
VDRC_ID 111003 is the "30D only" version of VDRC_ID 105838 (CG4236, Caf1)

RNAi collection extended – new shRNA lines added (Apr. 18, 2016)
Dear VDRC users,
The Vienna Drosophila Resource Center is pleased to announce a collection of new UAS-RNAi lines using short hairpin microRNA (shRNA) technology to extend and complement the current VDRC RNAi collection. Nearly 200 new lines are already available (www.vdrc.at) and a further 300 lines, currently in the VDRC production pipeline, will be made available to the community as soon as they are ready.
For commonly studied genes, where there is only 1 RNAi line in the VDRC GD or KK collection at present, we aim to add a further functional RNAi line to facilitate verification of phenotypes.
We have used shRNA technology to create these lines, as used extensively by the Transgenic RNAi Project (TRiP) and have collaborated with the TRiP team for hairpin design. The VDRC shRNA lines are consistent with, and complementary to, the TRiP shRNA lines, but the shRNAs target mRNA sequences which are independent of all current resources. The new collection therefore extends the range of RNAi lines available to the Drosophila research community.

New stocks added from Brennecke group (Feb. 18, 2016)
Dear VDRC users,
Our collection of "Other Resources" is growing and we have now added 175 stocks provided by Julius Brennecke (IMBA, Vienna). The new collection contains a variety of published stocks including 60 RNAi lines plus mutant alleles, tagged constructs, reporters, piRNA biogenesis reporters, miRNA sensors and tethering lines.
VDRC is open to donations of highly used stocks for integration into its community stock center collection. In addition, we offer a Private Stock Keeping Service where we will maintain and distribute your personal fly stock/plasmid collection on your behalf. VDRC host institute name change (Jan. 01, 2016)
Dear VDRC users,
As of 1st Jan 2016, the VDRC host institute will change its name from "Campus Science Support Facilities GmbH" (CSF) to "Vienna Biocenter Core Facilities GmbH" (VBCF).
All VDRC services will continue exactly as before, with the only change being that invoices will be issued by VBCF. All bank details remain unchanged.
